{"id":1491,"date":"2019-04-12T17:58:35","date_gmt":"2019-04-12T16:58:35","guid":{"rendered":"http:\/\/blogs.lshtm.ac.uk\/alsfordlab\/?p=1491"},"modified":"2019-06-04T16:18:06","modified_gmt":"2019-06-04T15:18:06","slug":"decoding-anti-leishmanial-drug-efficacy-determinants","status":"publish","type":"post","link":"https:\/\/blogs.lshtm.ac.uk\/alsfordlab\/2019\/04\/12\/decoding-anti-leishmanial-drug-efficacy-determinants\/","title":{"rendered":"Decoding anti-leishmanial drug efficacy determinants."},"content":{"rendered":"<p>After a couple of months on <a href=\"https:\/\/www.biorxiv.org\/content\/10.1101\/605873v2\" target=\"_blank\" rel=\"noopener\">BioRxiv<\/a> our first pre-print has been accepted for publication in <a href=\"https:\/\/aac.asm.org\/content\/early\/2019\/05\/29\/AAC.00795-19\" target=\"_blank\" rel=\"noopener\"><em>Antimicrobial Agents &amp; Chemotherapy<\/em><\/a>!<\/p>\n<p id=\"page-title\" class=\"highwire-cite-title\">&#8216;<span style=\"color: #0000ff\">Chemogenomic profiling of anti-leishmanial efficacy and resistance in the related kinetoplastid parasite <em>Trypanosoma brucei<\/em><\/span>&#8216;<\/p>\n<p>Selecting our <em>T. brucei<\/em> RNAi library in the anti-HAT drugs revolutionised our understanding of their mode of action and the ways the parasite can develop resistance. Unfortunately, an equivalent approach doesn&#8217;t yet exist in the related kinetoplastid parasite, <em>Leishmania<\/em>, which causes leishmaniasis, a devastating disease that&#8217;s endemic to a wide swathe of the tropics and sub-tropics.<\/p>\n<p>However, the <em>T. brucei<\/em> and <em>Leishmania<\/em> genomes exhibit remarkable synteny, and these parasites share much in their biochemistry and cell biology (though their life styles are different &#8211; extra and intracellular, respectively). Therefore, we decided to select the\u00a0<em>T. brucei<\/em> RNAi library in the anti-leishmanial drugs to see what insights we could gain into anti-leishmanial drug action and potential resistance mechanisms.<\/p>\n<p>We identified 44 proteins, including <em>T. brucei<\/em> orthologues of the two known <em>Leishmania<\/em> drug transporters. The remaining 42 hits hadn&#8217;t previously been associated with anti-leishmanial drug action. Several of these were validated in <em>T. brucei<\/em>, highlighting the importance of a lysosomal transporter to paromomycin, and membrane trafficking and structure to amphotericin-B, as well as identifying a potential contributor to amphotericin-B\/miltefosine cross-resistance.<\/p>\n<p>This cohort of proteins represents a valuable resource for future study in <em>Leishmania<\/em> and has revealed some intriguing biology in <em>T. brucei<\/em>.<\/p>\n","protected":false},"excerpt":{"rendered":"<p>After a couple of months on BioRxiv our first pre-print has been accepted for publication&#8230;<\/p>\n","protected":false},"author":116,"featured_media":0,"comment_status":"closed","ping_status":"closed","sticky":false,"template":"","format":"standard","meta":{"footnotes":""},"categories":[12450],"tags":[],"class_list":["post-1491","post","type-post","status-publish","format-standard","hentry","category-publication","odd"],"yoast_head":"<!-- This site is optimized with the Yoast SEO plugin v26.9 -
