{"id":454,"date":"2013-04-18T11:11:21","date_gmt":"2013-04-18T10:11:21","guid":{"rendered":"http:\/\/blogs.lshtm.ac.uk\/alsfordlab\/?p=454"},"modified":"2014-11-26T11:30:40","modified_gmt":"2014-11-26T10:30:40","slug":"speaking-at-kmcb-meeting-woods-hole-ma-april-2013","status":"publish","type":"post","link":"https:\/\/blogs.lshtm.ac.uk\/alsfordlab\/2013\/04\/18\/speaking-at-kmcb-meeting-woods-hole-ma-april-2013\/","title":{"rendered":"Speaking at KMCB meeting, Woods Hole MA, April 2013"},"content":{"rendered":"<p>The\u00a05th Kinetoplastid Molecular Cell Biology meeting in Woods Hole MA kicks off on the 21st April, and I&#8217;m going to be speaking about some of the findings from our latest RNAi library screen.<\/p>\n<p><strong>Inhibition of <em>Trypanosoma brucei<\/em> cathepsin-L increases sensitivity to lysis by human serum<\/strong>.<\/p>\n<p>Human serum (HS) is highly effective at killing non-human infective trypanosomes, such as <em>T. b. brucei<\/em>, <em>T. congolense<\/em> and <em>T. vivax<\/em>\u00a0(though, unfortunately, <em>T. b. gambiense<\/em> and <em>T. b. rhodesiense<\/em> are unaffected, explaining their ability to cause devastating disease in humans).<\/p>\n<p>From the work of several groups over the last two decades we know a huge amount about the mode of action of the HS\u00a0trypanolytic factor (TLF; <a title=\"PubMed\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/20398209\" target=\"_blank\">Vanhollebeke &amp; Pays, 2010<\/a>), as well as some of the ways that human infective African trypanosomes have evolved resistance (<a title=\"PubMed\" href=\"http:\/\/www.ncbi.nlm.nih.gov\/pubmed\/23059119\" target=\"_blank\">Stephens <em>et al<\/em>, 2012<\/a>). However, although it&#8217;s known that TLF enters the parasite <em>via<\/em> the haptoglobin-haemoglobin receptor (HpHbR), the proteins that enable its transit to the lysosome are a mystery.<\/p>\n<p>HS selection of the bloodstream form\u00a0<em>T. b. brucei<\/em> RNAi \u00a0library, and Illumina high thoughput sequencing of the remaining RNAi fragments, has identified proteins\u00a0that likely influence the transit of TLF to the trypanosome lysosome and its action therein.
